I have replication going from 1 device to another. The path is Device --> Switch --> Router --> Switch --> Device.
On the return, the device is sending: Device -->Switch --> Switch --> Device.
How does this affect traffic? Does it slow it down because its going a different way than what it was recieved?

Is device 1 on a different vlan than device 2 and if so if one of the Switches a layer 3 switch.

Different VLANS, but switches are 3750

Rob,
If the traffic is indeed coming the same way back then you would definately see some performance slow down as it will be asymmetric routing process. this will slow down the traffic process a little bit as the return traffic is not taking the same patch to the client.
