In a CVP 3.1 environment I have a single license file for 300 ports and just purchased an extra 150 ports.
My question is should I now have a single license file (for 450 ports) or can there be multiple license files (one for 300 ports and another for 150) in the C:\Cisco\CVP\Server\license directory?

Niall, should be pretty easy to test. App Admin will tell you in the logs how many ports you have.

I suspect you might run into HW limits if you have a single combo box, I hope these ports are spread among some, the VB limit is 400 ports.
