I would like to know if it's possible to assign single unity mailbox for multiple users (say around 100)?

The requirement is to setup a single mailbox for help desk users so that if a caller leaves a message any agent will be able to check it.

there are many ways to do this.
I would make the 2nd line on the phone for every help desk agent the same extension. (shared line) that is the help desk extension.
Configure that line (MWI Policy to light and prompt)
anyone else have a better idea?

if users do not have already their own VM box configure alternate extensions on one subscriber

I do not think you can do 100 alternate extensions.
