If you use a L2 access-layer connected to a L3 distribution layer then the vlans are defined on the distribution layer switches and propogated by VTP to the access-layer switches. The inter-vlan routing would take place on the distribution switches.
If you use a L3 access-layer then the vlans and L3 vlan interfaces are defined on the access-layer switches.
