We have about 20 servers with dual NICs, we want to run NIC teaming with port channel enable, so we need to create 20 port-channel interfaces. I want to know will this cause a performance issue in a 6509 switch with sup-720?

you should be fine:
>> With Release 12.2(18)SXE and later releases, the group_number can be 1 through 256, up to a maximum of 128 port-channel interfaces. With releases earlier than Release 12.2(18)SXE, the group_number can be 1 through 256, up to a maximum of 64 port-channel interfaces.

Are you planning on connecting your servers to 2 6509s?
We have our servers with NIC teaming connected to a pair of VSS switches running LACP. This way you get NIC redundency and switch redundency

I did not know about VSS until you mentioned. I think it is too much for us to implement that now. We will get the NIC redundancy only this time. But could answer my question, I have about 20 LACP in one 6500, is this no problem, or too many, I just what to get an idea?

Cisco IOS Release 12.2SX supports a maximum of 128 EtherChannels.

You should be fine with the number of port channel you need, but make sure you put them on different line cards or at least 2, so if one card goes bad you don't loose all your servers. Also, since you don't have a second switch, Sup redundancy may be a good idea.
