Is there a way to map a single Public IP address to
multiple private IP addresses through an ASA utilizing HTTP Host Header Records like a
Microsoft ISA Server does?

You will need a Load Balancer to do that - something like an ACE appliance from Cisco.
An ASA is just a stateful firewall which can do port forwarding but does not do deep packet inspection or inspect HTTP headers.
Do you want to replace the ISA server with an ASA firewall?

Yes, we are replacing an ISA with the ASA. We are just going to use a couple of new Public addresses to make the multiple Web Sites functional. We will also have to modify the DNS.

Yes using multiple public addresses (granted you have a few to spare) is the easiest option in that case.
