You really want one of the hosts to be dual stacked, so that the communication can be native v4 or native v6. The alternatives are very ugly and break most protocols that are more complicated than simple TCP connections.
Have the v6-only host talk to a NAT64 gateway - ASA 9.x firewalls might be able to do this - in preference to trying to having the v4-only host talk to a NAT46 gateway.
