I know that it's best practice not to use spanning-tree portfast and spanning-tree bdpuguard on trunk links between switches but is it advisable to use on trunks to VM hosts?

 You can turn on portfast  towards servers/VMs.

For Nexus it is:

spanning-tree port type edge trunk

They are not nexus switches they are 4506.

Should we enable portfast and or bdpuguard?

portfast yest the command on the 4500 is;

spanning-tree portfast trunk

Don't see a need for bpduguard.
