I have two sites data center runing on OTV, we use unicast, i have router1 and router 2 in site A, router 3 and router 4 in site B, router 3 and router 4 has configured adjancy server (router A ip, router B ip) under overlay interface, do I need to define adjancy server ip(router A's ip) in router B 's overlay interface? thanks

Correct, you would use the following configuration...

Device# configure terminal
Device(config)# interface overlay 1
Device(config-if)# otv use-adjacency-server 10.10.1.2 unicast-only
Device(config-if)# end
