Hi,
Please refer the digram a clarify few doubts;
1) Is the connection between router to router is necessary? What traffic is passing through that. The routing is doing by Switch.
2) Which layer should be the link between router and switch be ? (L2/L3).
3) I am using Etherchannel with Des - mac configuration, is that be good as the number of users are more (1500+).

What are the business objectives of design?
Based on the diagram
1) I would keep the router to router connection for redundancy.
2) I would make the connection between the router and the switch L3, that way you can control the routing. Assign the route to the switch priority over the WAN connection to the other router
3) Not to sure about this, I would increment the users little bit at a time and now put everyone on at once if possible and monitor your processes to make sure they can handle it

You can actually login to the switch and run a "test etherchannel" to see what the hash will come up with based on source and destination.
