Does the 4503 have to be powered down to install a new 48 port switch module?

No, you don't have to power down the switch to install/swap line cards. All line cards are hot swappable.

It indicates the backplane can handle data up to 64 Gbps and the forwarding throughput capacity is 48 million packets per second.

Layer 2 Hardware forwarding is 48Mpps
Layer 3 CE forwarding routing is also 48Mpps.

With 64Gbps switching fabric switch can handle upto 64Gbps traffic with a forwarding rate of 48 million packet per second. Forwarding rate is the rate at which the packets can be transmitted to the switch fabric for the processing.
