is it enough if one physical port is set to no switchport for making the intervlan communication to happen and should we need to enable ip routing command?

You'll still need to turn on ip routing.

Just to clarify u r trying to use a single port or multiple ports for intervaln routing by converting it/them to L3 port.if u r using a single port then i think it will not support sub-intefaces.
