Does it have to be configured for multicast to work? If multicast is enabled can receiver and sender not communicate over multicast routing with the RP?

Depends on the multicast routing protocol. PIM-DM and DMVRP don't used RPs.

Just to add to what Joseph said, source specific multicast (SSM) doesn't use RP either as the receiver already knows about the source and will send the joing directly to it (source tree rather than RP tree).
