Yup. Say for instance extension 1234 does not have a Unity mailbox. You could make a Call Handler with the DTMF ID (extension) of 1234 that is set to transfer to 1234.
Creating a bunch of call handlers for LOTS of numbers can be a pain, I realize... there's a tool on my web site to help with this in cases where you want hundreds of extensions to be accesible... it's call the Bulk Handler Creation tool (not real creative, but it works OK).
