Where is the delay? Right after they enter their password or when they get their message count or after you enter your ID but before you enter your password? Is it a delay when they hit their "messages" button till they hear a prompt? When you say sometimes, is this for particular accounts or does anyone and everyone get hit with a delay from time to time? How long is very long? 30 seconds?
Couple of common issues. Fragemented message stores can cause the message filters to take a long time (i.e. you'll get a dealy waiting for the message stack to return and give you counts on your totals). You can check the exchange logs to see if it's finishing the nightly defrags at least, we've had sites wire down the time this is allowed to run and it wasn't finishing at night which left folks at the end of the list high and dry as their boxes slowly fragmented badly...
Network traffic can be heavy/bandwidth can be tight which will cause delays in connecting to remote Exchange servers.
You may need to run the Exchange optimizer if you've added/removed a number of users (if you've added/removed 100+ users without optimizing it's probably a good idea regardless).
