by default,does 3550 need to define vlan in vlan database?

before creating vlan interface or assign a port to a vlan

Yes, you need to create VLAN using either config-vlan Mode or the VLAN databse. You can find more info on CCO at

You can only define 1-1005 in the vlan database. to define extended vlan range upto 4096, use the configuration mode vlan commands

You'll need the EMI image for Layer 3 features
