You might want to try using NBAR- network based app. recognition. its not supported in all router platforms. Needs CEF to be supported and enabled on the router. (typically starting from 2600, u can use NBAR). This feature helps in classification of traffic according to protocol type and collects statistics of each protocol flow.
ip nbar protocol-discovery is the command to be enabled under the router interface, and show ip nbar protocol-discovery should indicate the statistics of each protocol, 5 minute bit rate (bps) (both inbound and outbound) on that interface. You ll also get the packet count and byte count information for each protocol. Check out nbar feature for more details on what all protocols are supported. als new protocols can be supported by loading modules available from Cisco for download.

Or obtain a packet sniffer. I recommend CA's Sniffer Pro.
