I have two Catalyst 4507R with two Supervisor 4 installed for each.

To use both giga ports on board (in practice to connected this switch in channel with four links hence giga 1/1,1/2,2/1,2/2 available)

How have to I configure Supervisors ?

You would have to. I don't know what is more important to you....redundancy or more Gig ports? Please read this document.
