I have a Cisco 3640 that has a WIC-1DSU-56K4 and a 2621 that has a WIC-1DSU-56K4. What do I need to do; i.e. wiring, etc.; to test the configuration of these devices before I deploy the 2621 to my remote office?

I would think you could connect the dsu's back to back with a crossover cable (pins 1to7, 2to8). Make sure you set one side to clock internal and the other to clock from line for the test. Here's a link with sample back-back config.
