The answer is NO.
The spanning tree protocol is layer2 and the IP directed broadcast is layer 3.
When you put the "no ip directed-broadcast" under an interface it means that when a subnet broadcast arrives at the router's interface
the router will not route that broadcast to its subnet.
Remember there are applications that can produce subnet broadcast
The broadcast will look like this 192.168.1.255 for example and if the router has a route entry for this network and "ip directed broadcast" under the receiving interface then the packet will be routed. For security reasons, I advise you to make sure you have "no ip directed-broadcast" configured under the interface. From version 12.0 onwards, ip directed broadcast is disabled by default, that is, all IP directed broadcasts are dropped.
