WS-G6488 is a 10GBASE-LR Optical Interface Module (OIM). This OIM installs in a 1-port 10 Gigabit Ethernet Base Module, part number WS-X6502-10GE. The WS-X6502-10GE works with Supervisor Engines 1A, 2, and 720.
WS-XENPAK-LR is a 1-port 10GBASE-LR XENPAK (modular optical interface similar in concept to a GBIC). This XENPAK must be installed in either of the two non-blocking ports on the 2-port 10 Gigabit Ethernet Module, part number WS-X6802-10GE; or in any one of the ports on the high-density 4-port 10 Gigabit Ethernet Module, part number WS-X6704-10GE. Both the WS-X6802-10GE and WS-6704-10GE work ONLY with the Supervisor Engine 720, however.
The optics are the easy part: a WS-G6488 can be connected to a WS-XENPAK-LR using from 2m to 10km of single mode fiber and SC connectors. It's the rest that's complicated: what Supervisor Engine(s) you have, how you intend to use the 10 Gig interfaces in your network, etc.
