Is it possible to block a specific URL using Access lists, or do you have to use the IP?

yes you can do it with IOS Routers similar like blocking the "Code Red Worm". Look at http://www.cisco.com/warp/public/63/nbar_acl_codered.shtml#acl
Modify the class-map to your needs, build a policy-map and handle matching policies in the access-list.
