Hi,
I've a deployment of a TP1000 and a TP500. Does anybody know if the Telepresence Manager is mandatory to establish calls between this two endpoints?

The answer is in the SRND. Telepresence Manager is not mandatory.

No it is not. Setup as end points just like any other end point in CUCM they are able to make TP calls between them selves.
