Does route map works in layer 2 routing or switching devices...
is it a must to make sure all the interfaces must be in layer 3?

A route-map is used on a router or layer 3 switch.
You probably want to start by layer 2 switching, (how arp works), and then layer 3 routing.
After you get the routing piece figured, then worry about route-maps.
